Summary
Twelve patients with primary hyperlipoproteinemia type IIa according to Fredrickson were treated after a six week placebo period with a new cholesterol-lowering resin. Colestipol (U-26.597 A, Upjohn). The drug was given orally, five g three times daily. Ten patients completed the trial and were treated with Colestipol during one year. From the third month of treatment a significant decrease of their cholesterolemia was observed (P<0.05). This cholesterollowering effect persisted after twelve months with a mean decrease at the end of the trial of 16.6%. Serum triglycerides, glycemia and parameters of the calcium and phosphate metabolism were not influenced by the resin. A decrease in uricemia was noticed. The two patients with an abnormal oral Glucose Tolerance Test responded less or not at all. One patient interrupted the treatment due to gastro-intestinal complaints. The other patients did not show any untoward reactions except for episodes of moderate constipation. There were no signs of toxicity.

Douze patients souffrant d’hyperlipoprotéinémie essentielle (type IIa selon Fredrickson) ont reçu, après une période initiale de six semaines de placebo, une résine hypocholestérolémiante nouvelle, le Colestipol (U-26.597 A, Upjohn) à la dose de 5 g 3×/j. Dix patients ont terminé l’étude menée pendant douze mois. Une baisse significative de la cholestérolémie a été constatée dès le troisième mois (P<0.05). Au bout de douze mois la chute moyenne de la cholestérolémie était de 16,6%. La triglycéridémie, la glycémie et le métabolisme phospho-calcique n’étaient pas influencés par le traitement. Une baisse de l’uricémie a été constatée. Le traitement par le Colestipol était inefficace chez les deux patients présentant un diabète latent. Un patient a abandonné précocement le traitement à cause de plaintes digestives. Trois autres patients ont souffert d’épisodes de constipation. Nous n’avons pas constaté de signes de toxicité générale.

Twaalf patiënten met een primaire hyperlipoproteïnemie type IIa volgens Fredrickson werden, na een placeboperiode van zes weken, behandeld met een nieuw cholesterolverlagend hars, het Colestipol (U-26.597 A, Upjohn). De dosering bedroeg drie maal vijf gram per os per dag. Tien patiënten beëindigden het volledige behandelingsschema van twaalf maanden. Bij hen werd vanaf de derde behandelingsmaand een significante cholestcrolemiedaling vast-gesteld (P<0.05). Na twaalf maanden bedroeg de gemiddelde cholesterolemiedaling 16,6%. De triglyceridcmie, de glycémie en parameters van de calcium- en fosfaatstofwisseling werden niet beïnvloed. Een uricemiedaling werd vastgesteld. De twee patiënten met een gestoorde GTT beantwoordden weinig of niet aan de behandeling. Eén patient onderbrak voortijdig de therapie wegens gastrointestinale bezwaren. Bij de overige patiënten waren er op enkele episodes van obstipatie na gcen bijwerkingen. In zoverre dit met de toegepaste methode kan achterhaald worden, leken er zich geen tekenen van toxiciteit voor te doen tijdens de behandelingsperiode.
