The complete mitochondrial genome of Whitmania acranulata

Abstract

The complete mt genome sequence of Whitmania acranulata was obtained by PCR, containing 37 genes with 13 protein-coding genes, 22 transfer RNAs (tRNAs), and 2 ribosomal RNAs (rRNAs).

Morphological classification of Haemopidae is relatively difficult due to various kinds and similar shape. In this study, the complete mt genome of W. acranulata was sequenced.

Whitmania acranulata was captured in the pond in Anhui Province (31°53′10″N, 117°14′45″E). The specimen is stored in Fisheries Institute, Anhui Academy of Agriculture Sciences. We amplified mitochondrial DNA using PCR method as reported (Pan and Jiang Citation2018). The complete mitochondrial genome of W. acranulata is 14,468 bp (MK347500).

The genome of W. acranulata is encoded on heavy strand and in one direction. The genome contains a total of 37 genes, including 13 protein-coding genes, 22 transfer RNAs, and 2 ribosomal RNAs. The 13 protein-coding genes in the mt genome of W. acranulata also share similar features in start and stop codons with those in other Hirudiniformes (Shen et al. Citation2011; Ye et al. Citation2015). The start codon, ATG, is observed for cox1, cob, atp6, nad2, nad3, nad4, nad4L, nad5 and nad6, and ATT observed for atp8 and nad1, and GTG, ATA for cox2, cox3, respectively. Five genes use TAA as complete stop codons (nad4L, nad5, atp6, cob, and cox2), whereas other seven genes appear to end in incomplete stop codons with T––.

Phylogenetic analysis was performed using 13 protein-coding genes by neighbor-joining (NJ). The clade containing Hirudinea and Oligochaeta is well revealed with high supporting value in the phylogenetic tree ().
