Abstract

The complete mitochondrial genome sequence of Metapenaeus ensis was obtained by PCR, containing 37 genes with 13 protein-coding genes, 22 transfer RNAs (tRNAs), two ribosomal RNAs (rRNAs) and a non-coding control region.

Morphological classification of shrimp in Penaeidae is relatively difficult due to various kinds and similar shape. In this study, the complete mitochondrial genome of Metapenaeus ensis was sequenced.

Metapenaeus ensis was captured in East China Sea near Wenzhou City (28°15′18.69″N, 121°9′16.96″E) and stored in Zhejiang Mariculture Research Institute. We amplified mitochondrial DNA using PCR method (Pan and Jiang Citation2018). The complete mitochondrial genome of M. ensis is 15,956bp (MK500697).

The mitochondrial genome of M. ensis are encoded on two strands and in two directions. It totally contains 37 genes, including 13 protein-coding genes, 22 transfer RNAs and two ribosomal RNAs. The 13 protein-coding genes share features in start and stop codons with those in other Penaeidae shrimps (Mao et al. Citation2016; Zhong et al. Citation2018).

Phylogenetic analysis was performed using 13 protein-coding genes by neighbour-joining (NJ). The clade containing Metapenaeus, Parapenaeopsis, Penaeus, Fenneropenaeus and Marsupenaeus is well revealed with high supporting value in the phylogenetic tree. It is indicated that Fenneropenaeus, Penaeus and Marsupenaeus are closely related, forming a clade, with Metapenaeus, and Parapenaeopsis as a sister group ().

Figure 1. Neighbour-joining phylogenetic tree inferred from amino acid sequence dataset of 13 protein-coding genes for 11 Penaeidae mitochondrial genomes. The tree shows the topology based on concatenated data of 13 mitochondrial encoded protein sequences (nad2, cox1, cox2, atp8, atp6, cox3, nad3, nad5, nad4, nad4L, nad6, cob, and nad1). Reconstruction was performed by MEGA X (64-bit). Values at nodes are proportions of 1000 bootstrap pesudorelicates in which the nodes were recovered.
