DETERMINATION OF FLEROXACIN AND SPARFLOXACIN SIMULTANEOUSLY BY TLC-FLUORESCENCE SCANNING DENSITOMETRY

Abstract

A new sensitive, fast thin layer chromatography-fluorescence scanning densitometry (TLC-F) method is described for the simultaneous determination of Fleroxacin (FLX) and Sparfloxacin (SPLX) using a mobile phase of ethanol : ethyl acetate : 1,2-dichloroethane : 10% aqueous ammonia (40 : 30 : 20 : 10 v/v) and a silica gel thin layer. Previously treated with 0.27 mol L−1 sodium edetate solution of pH 7.0, FLX and SPLX were separated from each other. The R f values are 0.4 and 0.59 for FLX and SPLX, respectively. The fluoresecent spots were scanned with a spectrodensitometer using 285 nm as the excitation wavelength and cut off filter of 400 nm. The linear ranges are 0.5–85 ng (FLX) and 0.5–100 ng (SPLX). The recovery of them in serum and urine is 95.6–102.0%. The relative standard deviations are <5.2%.
