Abstract

The matrix interference subjected by R‐ and S‐1‐phenyl‐1‐butanol in the biphasic Saccharomyces cerevisiae CCRC 21443 mediated culture cannot be eliminated by chiral high‐performance liquid chromatography (HPLC) with a single column of either Chiralcel OB or Chiralpak AD. The use of a single column also results in a long analysis time. The application of the column switching technique can hyphenate the Chiralcel OB column and the Chiralpak AD column together, to resolve not only the matrix interference, but a reduction of the analysis time for these chiral products in a complex medium. This coupled column system has an accuracy greater than 98.6% and a high precision of less than 1.6% relative standard deviation (RSD). The developed technique for on‐line sample clean‐up and analysis of phenyl‐n‐propyl ketone, R‐ and S‐1‐phenyl‐1‐butanol in the culture is successful.
