Determination of Urinary Metabolites of Coumarin in Human Urine by HPLC

Abstract

Liquid chromatography with fluorometric detection has been developed for the determination of the metabolite of coumarin (7‐hydroxycoumarin) in human urine. 7‐Hydroxycoumarin and 5,7‐dimethylcoumarin were separated simultaneously on a 25 cm × 4.6 mm i.d. Phenomenex CN 5 µm column with acetonitrile–water (40:60, v/v) as mobile phase at 1.0 mL/min, and detected at excitation 325 nm and emission 470 nm. The limit of detection was 0.28 ng/mL for 7‐hydroxycoumarin. Analysis of human urine 1–6 days after ingestion of oral Chinese medicines, led to the conclusion that the concentration of 7‐hydroxycoumarin was higher than for control urine.
