HPLC Method for the Pharmacokinetics Study of Acemetacin in Human Plasma

Abstract

A reproducible and specific high performance liquid chromatography (HPLC) method was developed to quantity plasma levels of acemetacin and its metabolite, indometacin in order to conduct a pharmacokinetics study. Plasma samples were prepared by liquid‐liquid extraction using ethyl acetate. Extracted samples were analyzed on a reverse‐phase C18 column with a mobile phase consisting of 20 mM phosphate buffer (pH 2.9) and acetonitrile (60:40, v/v) and UV detection at 254 nm. Using this method, the pharmacokinetics study was carried out on eight volunteers after a single oral administration.
