Effects of Low-Dose Lynestrenol as a Contraceptive

Summary

The effects of lynestrenol, 0.5 mg. per day, administered continuously as an oral contraceptive, were studied in 50 women over 1,146 menstrual cycles; 45 patients completed 24 cycles. No pregnancies occurred.

Subjective side-effects were not reported and variations in weight or blood pressure were not observed. No disturbances were found in various laboratory tests or in cytologic studies. Interference with the normal hormonal transformation of the endometrium was found in only 16 % of the biopsies performed. Menstrual bleeding lasted on average 4 to 5 days. Amenorrhoea was present in 0.4% of the cycles. Irregular bleeding (either spotting or breakthrough bleeding) was present in 14 % of the cycles: 6 patients (12%) stopped the treatment because of this.

It is concluded that the preparation studied is effective and very well tolerated. Patients should however be forewarned that cycle disturbances can take place, but other side-effects are minimal and the contraceptive efficacy is very high. This reassurance will reduce the drop-out rate.
