Biosynthetic engineering of polyketide synthases

Abstract

Polyketides are a diverse class of natural products with commercial significance for the pharmaceutical and agrochemical industries. Rapid advances have been made in the understanding of polyketide biosynthesis. It is now possible to modify the biosynthetic machinery to produce radically altered structural analogues of the parent polyketides that are not readily accessible by conventional technologies, such as total- or semi-synthesis. This review summarises some of the published patents and patent applications in this relatively new field, focusing both on the core technological issues governing success in this area and on specific examples of patents describing the use of these technologies to produce molecules of therapeutic significance.
