Novelty: Methods for mapping discrete DNA sequences complementary to regions of a eukaryotic chromosome and for isolating a gene of interest are disclosed.
Biology: Methods for ordering a set of genomic clones complementary to a region of a eukaryotic chromosome are provided. The linear order of clones from a chromosome-specific library is determined by in situ hybridization, the clones are then analysed for the presence of rare cutter restriction enzyme sites and for restriction fragment length polymorphism. This method substantially reduces the number of clones which must be analysed to define markers of appropriate spacing. Specific genes of interest may be isolated by identifying candidate genomic clones which contain a rare cutter site and have a map position near the locus identified previously by genetic mapping.
