Abstract
Novelty: A method for treating or preventing neuronal cell damage is claimed. It involves the iv administration of a neurotrophic factor which can be GFGF, aFGF, CNTF, BONF, NGF, NT3, NT4, IGF-1 or IGF-I1. The claimed uses include cerebral ischaemia, trauma, neurodegeneration or cardiac surgery.
Biology: In the mouse MCA model of focal ischaemia, a dose of GFGF or NGF was given 15 minutes after ischaemia and then the same dos'e one a day for seven days. The dose of GFGF needed to give a significant reduction in damage over placebo was 150 pg and for NGF was 565 pg. Data are also given for the rat four vessel occlusion model of global ischaemia and a gerbil global ischaemia model.
Chemistry: No chemistry is described. These neurotrophic factors are well reported and include methods of isolation and purification as well as recombinant technology for the production of the human neurotrophic factors.
