Patent Evaluation: Aminosterol antibiotic

Summary

Novelty: A pure aminosterol antibiotic called squalamine which can be isolated from the tissues of the common dogfish shark (Squalus acanthius) is claimed. The antibiotic is stated to be active against Gram-negative and -positive bacteria, fungi and protozoa.

Biology: The antimicrobial activity of squalamine was assayed in vitro against E. coli, S. aureus, P. aeruginosa and S. faecalis. Full results are presented in a table.

Chemistry: The isolation and characterization of squalamine is fully described. The determination of the tissue distribution is also described. Squalamine is (3β(N-[3-aminopropyl]-1,4-butanediamine)-7α,24-dihydroxy-5α-cholestane 24α-sulfate).
