Summary
Novelty: DNase treatment may be used in conjunction with accepted gene therapy (pulmonary administration of DNA-containing liposomes) for the treatment of cystic fibrosis. It is suggested that DNase will reduce cystic fibrosis mucus viscosity and enhance the efficiency of liposome-mediated gene delivery.
Biology: Cationic liposomes were prepared and their ability to deliver fluorescent lipid to cell membranes determined. Liposomes were also used to deliver cholera toxin subunit A to the cytoplasm of CEM cells. Membrane fusion of liposomes with CV-1 cells was inhibited by the presence of calf thymus DNA but was relieved by pre-treatment with DNase.
