Determination of the Sedimentation Coefficients of D-Glucose-6-Phosphate Dehydrogenase from Normal and Deficient Human Erythrocytes by S-Zonal Ultracentrifugation

Abstract

The sedimentation coefficients of D-glucose-6-phosphate dehydrogenase from normal and deficient human erythrocytes have been studied by s-zonal ultracentrifugation. Procedures for s-zonal ultracentrifugation of D-glucose-6-phosphate dehydrogenase after a DEAE-cellulose chromatography and of crude hemolysate have been developed. Equivalent sedimentation coefficients of 6.4 S have been found for normal as well as for deficient D-giucoss-6-phosphate dehydrogenase. The risk of overloading the gradient in s-zonal ultracentrifugation has been studied. Experimental evidence suggests that the best practical criterion in the assessment of overloading is dislocation of a standard substance.
