Kinetics of the Reaction Between DNP-Insulin and Homologous Antibody

Abstract

Kinetic and equilibrium studies were performed on the reaction between monovalent DNP-insulin (DNP-I) and antibodies specific for the 2,4-dinitrophenyl (DNP) determinant. The average intrinsic association constant as determined by fluorescence quenching was 1.2 × 107 M-1. Kinetic experiments, using a stopped-flow technique, revealed a biphasic behaviour, and the rate constant of association, obtained fran the initial linear portion of the rate data, was calculated to be 4.7 ± 0.5 × 106 M-1sec-1.
