Abstract
Paraffin sections from tissues fixed in nonneutralized 10% formalin or Carnoy's fluid were treated with saturated solutions of picric acid in each of the following solvents: water, 80%, 90%, 95% and absolute ethanol, and with half-saturated solutions in 95% ethanol for 5 min. Tests with the supernatant fluid 30-60 min after mixing the solid and the solvent showed that adequate concentration had not yet occurred; hence more time is needed, with 15 hr recommended. Only saturated solutions of picric acid in absolute and 95% ethanol removed all acid hematin-type pigment. The effectiveness of saturated solutions decreased with decreasing alcohol concentration.
