Arthritis of Rats Inoculated with Granuloma Pouch Exudate

Summary

Arthritis can be induced in the rat by treatment with the exudate formed in the granuloma pouch. The authors injected the exudate obtained from the 6-day granuloma pouch (exudate I) under the skin of the sole of rats. In another series of experiments exudate I was passaged in the granuloma pouch and the exudate thus obtained (called exudate II) was injected into the experimental animals in the same way. The swelling of the limb soon following the injection of both kinds of exudate has been ascribed to histamine action. By the intraperitoneal injection of phenergan the development of swelling could be delayed and its measure could be diminished.

Exudate II caused a more marked swelling of the ankle and paw, that persisted more stably during the experimental period (41 days) than in the case of the rats treated with exudate I. Histological study has shown inflammation spreading from the periarticular tissue toward the synovia, and in many cases synovitis could be observed.

La polyarthrite chronique évolutive peut être communiquée au rat traité par l'exsudat secrété dans la poche du granulome. Les auteurs ont injecté l'exsudat obtenu dans une poche de granulome de 6 jours (exsudat I) sous la peau de la plante de la patte chez des rats. Dans une autre série d'expériences, l'exsudat I a effectué un passage dans la poche du granulome et l'exsudat ainsi obtenu (nommé exsudat II) a été injecté chez les animaux en expérience par le même procédé. L'enflure du membre suivant de près l'injection des deux variétés d'exsudat a été attribuée a une action histaminique. L'injection intrapéritonéale de phénergan a retardé le développement de l'enflure et diminué ses dimensions.

L'exsudat II a provoqué une enflure plus marquée de la hanche et de la patte, qui a persisté plus longuement durant la période expérimentale (41 jours) que chez les rats traités au moyen d'exsudat I. L'examen histologique a révélé une inflammation s'étendant des tissus périarticulaires vers la synovie, et dans plusieurs cas on put observer une synovite.

Zusammenfassung

Arthritis kann der Ratte durch Behandlung mit dem in der Granulomtasche gebildeten Exsudat induziert werden. Die Verfasser injizierten das aus der 6 Tage alten Granulomtasche gewonnene Exsudat (exsudat I) bei Ratten unter die Haut der Fussohlen. In einer anderen Versuchsreihe wurde das Exsudat I in die Granulomtasche überführt und das so gewonnene Exsudat (Exsudat II) wurde den Versuchstieren auf dieselbe Weise injiziert. Das den Injektionen mit jeder der zwei Arten von Exsudat bald nachfolgende Anschwellen der Gliedmasse wurde der Histaminwirkung zugeschrieben. Bei der intraperitonealen Injizierung von Phenergan konnte die Geschwulstentwicklung verzögert und ihr Ausmass verringert werden.

Das Exsudat II verursachte ein auffallendes Anschwellen des Fussknöchels und der Pfote, das während der Versuchsperiode (41 Tage) länger anhielt als bei den mit Exsudat I behandelten Ratten. Eine histologische Untersuchung hat eine Entzündung gezeigt, die sich vom periartikulären Gewebe aus über die Synovia ausbreitete. In vielen Fällen konnte Synovitis beobachtet werden.
