The effect of induced defecation in honey bee queens on the number and viability of spermatozoa in the spermatheca after instrumental insemination

Summary

This study determined the number and viability of spermatozoa accumulated in the spermathecae of honey bee (Apis mellifera queens which did or did not defecate prior to instrumental insemination. The study used 64 samples of semen from the spermathecae of 33 defecating queens and 31 queens that did not defecate. Spermatozoa counts were estimated by computer-assisted sperm analysis with an HTM IVOS analyser. The viability of spermatozoa was determined by fluorescence staining with SYBR-14/propidium iodide (IP) and flow cytometry. Defecation by queens was found to have no effect on the number of spermatozoa accumulated in the spermatheca, but it did affect their viability.

Resumen

Este estudio determinó el número y la viabilidad de los espermatozoides acumulados en la espermateca de las abejas reina (Apis mellifera) que defecaron o no antes de la inseminación instrumental. Se utilizaron 64 muestras de semen procedentes de la espermateca de 33 reinas que defecaron y de 31 reinas que no defecaron. El número de espermatozoides se estimó por medio de un análisis computarizado de esperma con el programa HTM IVOS. La viabilidad de los espermatozoides se determinó por medio de la fluorescencia de tinción con yoduro de propidium (IP)/SYBR-14 y citometría de flujo. No se encontró efecto de la defecación de las reinas sobre el número de espermatozoides almacenados en la espermateca, pero sí afectaron su viabilidad.
