ABSTRACT
Fatty acid amide hydrolase (FAAH) degrades the endogenous endocannabinoid (eCB) anandamide and might be involved in the response to suggestions of analgesia in subjects with high hypnotizability scores (highs). Since the A allele of the FAAH C385A polymorphism (rs324420) is associated with lower FAAH activity, it was studied in 21 highs, 66 low hypnotizable individuals (lows), and 172 individuals not selected for hypnotizability (controls) representing the general population. No significant difference was observed among groups, but the A allele frequency showed a significant trend to increase from lows to controls and from controls to highs. Since eCB small differences can be amplified by eCB interactions with other neurotransmitters, a contribution of the FAAH polymorphism to the highs’ analgesia should not be excluded.

Abstract: Die Fettsäureamid-Hydrolase (FAAH) baut das endogene Endocannabinoid (eCB) zu Anandamid ab und könnte in die Antwort auf Suggestionen von Analgesie bei Probanden mit hohen Hypnotisierbarkeitsleveln (highs) involviert sein. Da die A Allele der FAAH C385A Polymorphismen (rs324420) mit niedrigerer FAAH-Aktivität assoziiert sind, wurden diese in 21 hoch-hypnotisierbaren, 66 niedrig-hypnotisierbaren Individuen (lows) und 172 Teilnehmern, die nicht für die Hypnotisierbarkeit ausgewählt wurden (controls) und die allgemeine Bevölkerung repräsentieren, untersucht. Es wurde kein signifikanter Unterschied in den Gruppen gefunden, aber die A Allel-Frequenz zeigte einen signifikanten Trend der Zunahme von den niedrig-hypnotisierbaren zu den Kontrollpersonen und von den Kontrollpersonen zu den hoch-hypnotisierbaren. Da kleine eCB-Unterschiede durch eCB-Interaktionen mit anderen Neurotransmittern verstärkt werden können, sollte ein Beitrag des FAAH-Polymorphismus bei der Analgesie in hoch-hypnotisierbaren nicht ausgeschlossen werden.

Résumé : L’hydrolase des amides d’acides gras (FAAH) dégrade l'anandamide cannabinoïde endogène (ACe) et pourrait jouer un rôle dans la réponse aux suggestions d'analgésie chez les sujets ayant des scores de susceptibilité hypnotique élevés (sujets très sensibles à l'hypnose (TSH)). Étant donné que l'allèle A du polymorphisme FAAH C385A (rs324420) est associé à une activité plus faible de la FAAH, il a été étudié auprès de 21 personnes TSH, 66 personnes peu sensible à l'hypnose (PSH) et 172 personnes non sélectionnées pour leur sensibilité hypnotique (groupe témoin) représentant la population en général. Aucune différence significative n'a été observée entre les groupes, mais la fréquence d’allèle A a eu tendance à évoluer à la hausse depuis le groupe des PSH jusqu’au groupe témoin, et depuis le groupe témoin jusqu’au groupe TSH. Étant donné que les petites différences ACe peuvent être amplifiées par les interactions de celui-ci avec d'autres neurotransmetteurs, une contribution du polymorphisme FAAH à l'analgésie des TSH ne devrait pas être exclue.

Resumen: La amida hidrolasa de ácidos grasos (FAAH por sus siglas en inglés) degrada la anandamida endocannabinoide (eCB por sus siglas en inglés) endógena y podría estar involucrada en la respuesta a las sugestiones de analgesia en sujetos altamente hipnotizables (altos). Considerante que el alelo A del polimorfismo FAAH C385A (rs324420) está asociado con menor actividad FAAH, se estudio 21 altos, 66 individuos poco hipnotizables (bajos) y 172 individuos que no fueron seleccionados por su hipnotizabilidad (controles) que representaban la población general. No se observaron diferencias significativas entre grupos, pero el alelo A mostró una tendencia significativa a incrementar entre bajos y controles y entre controles y altos. Como las pequeñas diferencias en eCB pueden amplificarse a través de interacciones entre eCB y otros neurotransmisores, no debería excluirse una contribución del polimorfismo FAAH en la analgesia de los altos.
