ABSTRACT
The present experiment was conducted to determine the expression kinetics of HSP70, HSP90, HSP110, HSF1, SOD, SOCS1, TLR2, TLR4, IL2, IL6, iNOS and eNOS in PBMCs exposed to higher temperature (42°C) in in-vitro culture system. PBMCs were exposed to elevated temperature of 42°C at 0, 1, 2, 4, 8, 12, 16, 20, 24 hr intervals and recovery at 37°C. The time course expression pattern of the heat stress induces genes were examined. The relative mRNA expression was significantly higher for iHSP70, HSP90, and HSF1 (p < 0.05) at 4 and 16 hr, for HSP110 at 8 hr, for SOCS at 12 and 24 hr whereas IL2, IL6, TLR2, TLR4, iNOS and eNOS showed significantly higher expression at 4 h of heat exposure. In the recovery study, HSP70 and HSP90, HSF1, eNOS, iNOS, TLR2 showed significant higher expression (p < 0.05) at 2 hr compared to control. There exists a negative correlation between HSP70 and all inflammatory cytokines. In conclusion, our findings suggest that at higher environmental temperature, interaction among HSPs, TLRs and cytokines possibly play a significant role as robust, intrinsic ameliorative mechanism to protect the animal from deleterious effect of thermal stress.
