ABSTRACT
Loading of Ferula assa-foetida essential oil on PLGA nanoparticles for improve its biological activity.
FAFEO-PNP were produced by W1/O/W2 method and after characterization, its antibacterial (Disc Diffusion, MIC, MBC), antioxidant (ABTS, DPPH, FRAP), pro-apoptotic (MTT, qPCR, AO/PI staining, Flow cytometery) and anti-angiogenic (CAM) effects were investigated. Cancer induction was performed to evaluate the antitumor effects of FAFEO-PNP.
FAFEO-PNP (size: 203.62 nm, PDI: 0.26 and ζ-p: -32.92 mV) inhibited HepG2 (IC50: 57μg/mL) cells. FAFEO-PNP induced the cell death by up-regulation of P53, TNF-α, P-21, Cas-3 and 9 genes. The activity of inhibiting free radicals and inhibiting the growth of different strains of bacteria in the presence of nanoparticles showed the antioxidant and antibacterial power of FAFEO-PNP. In vivo analysis including CAM and cancer induction assay confirmed the anti-angiogenic (reduction of vascular parametric factors) and anti-tumor effects of FAFEO-PNP. FAFEO-PNP can be considered as an anti-cancer agent in cancer therapy.
