Figure 1. Irisin, secreted abundantly by muscles during exercise, regulates bone resorption, and thermogenic gene program. Osteocytes and adipose cells express high level of αV/β5 integrins, a receptor of irisin. Irisin activation of αV/β5 in osteocytes promotes bone resorption by increasing RANKL and sclerostin expression. In addition, irisin-αV/β5 integrin signalling upregulates the expression of thermogenic genes (Ucp1 and Diol) in adipose tissues, which is responsible for fat expenditure.
