Abstract
With the increasing interest and use of messenger RNA (mRNA) for identifying body fluids in forensic science, appropriate controls need to be developed. An amplification positive control is an essential requirement for all PCR amplifications currently used in forensic biology, such as DNA profiling. For mRNA identification of body fluids, this means a control that amplifies all the relevant body fluids detected in an mRNA multiplex assay. We have developed an amplification positive control for mRNA identification of those body fluids commonly encountered in forensic science (semen, blood, menstrual blood, saliva and vaginal material). RNA from prostate, testes, peripheral blood leukocytes, endometrial cells, tongue and from vaginal swabs were combined, cDNA synthesized and then amplified to form an amplification positive control. Amplification of this positive control is reproducible between amplifications, when new cDNA is synthesized and used as an input template and when new lots of RNA are used for synthesizing the cDNA. This amplification positive control can be adapted for use with different mRNA multiplex assays.

Avec l’intérêt grandissant et l’utilisation croissante de l’ARN messager (ARNm) pour l’identification de fluides corporels en science médico-légale, des contrôles appropriés doivent être développés. Un contrôle positif d’amplification est une composante essentielle pour toutes réactions d’amplification PCR actuellement utilisées en biologie médico-légale tel le profilage ADN. Pour l’identification des fluides corporels, cela signifie un contrôle qui détecte les ARNm de tous les fluides corporels pertinents amplifiés lors d’une réaction multiplexe. Nous avons développé un contrôle positif d’amplification pour l’identification par l’ARNm des fluides corporels couramment rencontrés en science médico-légale (sperme, sang, sang menstruel, salive et le matériel vaginal). De l’ARN provenant de la prostate, de testicules, de leucocytes du sang périphérique, de cellules de l’endomètre, de la langue et de prélèvements vaginaux ont été combinés, l’ADNc a été synthétisé et ensuite amplifié pour former un contrôle positif d’amplification. L’amplification de ce contrôle positif est reproductible entre amplifications lorsqu’un nouveau ADNc est synthétisé et utilisé comme matériel de départ et aussi quand de nouveaux lots d’ARNm sont utilisés pour la synthèse de l’ADNc. Ce contrôle positif d’amplification peut être adapté pour une utilisation avec différents tests multiplexes d’ARNm.
