Effects of dietary calcium levels on development, haemolymph and antioxidant status of honey bee (Apis mellifera) larva reared in vitro

Abstract

In order to find out optimal dietary calcium levels for honey bee larvae reared in vitro, we studied the effects of larvae dietary calcium levels on the haemolymph and antioxidant status, as well as growth and development (weight, pupation and emergence rate) of larvae reared in vitro in different growth stages. Eight diet groups were chosen and one group (control) was fed a basal diet containing 43.99 mg/kg calcium; the other seven groups were fed diets with calcium levels ranging from 93.99 to 393.99 mg/kg. Calcium concentration of the haemolymph of larvae significantly (p < .001) increased with increased dietary calcium. When the dietary calcium levels ranged from 193.99 to 243.99 mg/kg, larvae, pupae and emerged bees had the highest weight, and larvae and pupae had a significantly (p < .05) higher total antioxidative capacity. With calcium from 93.99 to 243.99 mg/kg, larvae had a significantly (p < .001) higher pupation rate. Larvae fed diets with calcium levels at 143.99 and 193.99 mg/kg had a significantly higher emergence rate (p < .001). The lowest malondialdehyde concentration of larvae fed 93.99 mg/kg calcium was .19 mmol/ml, while that of pupae 143.99 mg/kg calcium was .15 mmol/ml. We conclude, therefore, that larval dietary calcium levels from 143.99 to 243.99 mg/kg are optimal for larval growth and physiological function.

Con el fin de encontrar los niveles óptimos de calcio en la dieta para la cría in vitro de las larvas de abeja de la miel, estudiamos los efectos de los niveles de calcio en dieta sobre la hemolinfa y el estado antioxidante, así como sobre el crecimiento y el desarrollo (peso, pupación y tasa de emergencia) de las larvas criadas in vitro en diferentes etapas de crecimiento. Se seleccionaron ocho grupos de dietas, y un grupo (control) fue alimentado con una dieta basal conteniendo 43,99 mg/kg de calcio; los otros siete grupos fueron alimentados con dietas con niveles de calcio que fueron desde 93,99 hasta 393,99 mg/kg. La concentración de calcio en la hemolinfa de las larvas aumentó significativamente (P < 0,001) con el aumento de calcio en la dieta. Cuando los niveles de calcio en dieta oscilaron entre 193,99 y 243,99 mg/kg, las larvas, las pupas y las abejas emergidas tuvieron el mayor peso, y las larvas y pupas presentaron una capacidad antioxidante total (T-AOC) significativamente superior (P < 0,05). Con niveles de calcio de 93,99 a 243,99 mg/kg, las larvas mostraron una tasa de pupación significativamente superior (P < 0,001). Las larvas alimentadas con dietas con niveles de calcio en 143,99 y 193,99 mg/kg tuvieron una tasa de emergencia significativamente mayor ( < 0,001). La menor concentración de MDA en larvas alimentadas con 93,99 mg/kg de calcio fue de 0,19 mmol/ml, mientras que la de las pupas a 143,99 mg/kg de calcio fue de 0,15 mmol/ml. Se concluye, por tanto, que niveles de calcio en la dieta de las larvas entre 143,99 y 243,99 mg/kg son óptimos para el crecimiento larval y la función fisiológica.
