Bloat in cattle 45. Identification of gland sources of bovine salivary proteins of bovine salivary proteins

Abstract

Parotid, mandibular, sublingual, ventral buccal, intermediate buccal, dorsal buccal, pharyngeal glands, epiglottis, and soft palate tissue were excised from 10 cattle of known susceptibility to bloat. Salivary proteins were extracted from the glands and identified using immunoelectrophoretic methods and ultracentrifugation. Parotid and ventral buccal glands contained electrophoretically defined protein Bands 2,3,4,5,6,7, 10, and lOs. Mandibular extracts contained Bands 2,3,6,7, and 8 together with mucoprotein. Sublingual and pharyngeal glands contained Band 9 protein and mucin. Intermediate and dorsal buccal glands contained low concentrations of all the low molecular weight proteins and mucin. Glands of the epiglottis and soft palate contained mucin, but no low molecular weight proteins. Immunocytochemistry showed that all the low molecular weight proteins were associated with serous and not mucous secretory cells. Band 4 and 10 proteins were present in the proteoserous cells of secretory endpieces of the parotid and ventral buccal glands and in the demilune cells of the intermediate and dorsal buccal glands. Band 8 and 9 proteins were localised in the demilune cells of the mandibular and sublingual glands respectively.
