Feature Article: The Design of Columns for Gel Permeation Chromatography in High Throughput Applications

Abstract

An investigation was performed to determine the optimum parameters for GPC columns to be used in high throughput analysis and combinatorial materials discovery applications. The influence of column packing, column dimensions, and linear flow rate through the column on the specific resolution of GPC experiments was assessed. It was found that maximizing the pore volume of the packing material increased resolution but did not affect the speed of the analysis while either reducing the column length or increasing the linear flow rate through the column reduced resolution but at a significant gain in speed and therefore sample throughput. Increasing the internal diameter of the columns (maintaining identical linear flow velocity) did not affect the resolution or analysis speed but did consume more solvent. Based on these findings, the PL Rapide column range was developed for rapid GPC to maximize throughput with minimal impact on resolution.
