Abstract
A technique is described by which the mycelial structures of a dermatophyte attacking a hair in vitro, which are embedded within the hair and therefore difficult to observe, can be studied in detail. On soaking the attacked hair in 20% KOH the keratinaceous material is dissolved and the freed mycelial structures can be examined in unstained wet mounts by phase contrast microscopy. By this means it was confirmed that the great majoity of perforating organs of Nannizzia persicolor and Microsporum canis originate from eroding fronds, but occasionally a perforating organ is produced from a normal aerial hypha. The development of perforating organs in human hair is traced and the outgrowths from them, which are responsible for the destruction of the medulla and cortex of the hair, are described.
