Human lymphocyte blastogenesis induced by living and dead Histoplasma capsulatum yeasts and soluble yeast autolysate

Abstract

Human peripheral blood mononuclear cells were cultured with either replicating yeast phase Histoplasma capsulatum, intact dead yeasts or soluble yeast phase autolysate antigen for induction of lymphocyte blastogenesis. H. capsulatum replicated to the same extent in cultures of lymphocytes from histoplasmin skin test-positive and negative cell donors, but stimulated far greater blastogenesis in the former. Dead H. capsulatum yeasts, which were more easily quantified, caused nearly as much specific blastogenesis as live yeasts. Blastogenesis was induced by as few as 1 dead yeast per 10,000 lymphocytes, indicating the antigenic potency of Histoplasma yeasts. Specific blastogenesis was also caused by soluble yeast autolysate. Thus, replicating H. capsulatum yeasts, dead yeasts and autolytically released soluble yeast phase antigen each stimulated specific blastogenesis of lymphocytes from persons sensitized to H. capsulatum in vivo.

Zusammenfassung

Aus venösen Blutentnahmen gewonnene Monozyten und Lymphozyten wurden in Gegenwart der Antigene von proliferierenden oder abgetöteten intakten Hefezellen oder von Hefezellen-Autolysat (Histoplasma capsulatum) inkubiert zur Induktion der Lymphozytenblastogenese. Die Proliferationsrate der intakten Hefen zeigten keinen Unterschied in den Kulturen von Spendern mit positivem oder negativem Histoplasmin-Hauttest. Die Blastogenese der Lymphozyten dagegen war wesentlich ehöht in den Kulturen von positiven Spendern. Abgetötete H. capsulatum Hefen erhöhten die spezifische Blastogenese fast ebenso gut wie die proliferierenden Hefezellen, wobei die abgetöteten Organismen einfacher zu verwenden waren. Zur Veranschaulichung der antigenen Potenz der H. capsulatum Hefen konnte gezeigt werden, dass die Blastogenese mit einer totan Hefezelle per 10,000 Lymphozyten induzierbar war. Die spezifische Blastogenese war auch vorhanden nach Inkubation mit Hefen-Autolysat. Es konnte daher gezeigt werden, dass die Antigene von proliferierenden oder abgetöteten intakten Hefezellen oder von Hefen-Autolysat (H. capsulatum) in der Large waren, die spezifische Blastogenese in Lymphozyten von Spendern zu induzieren, die vorher in vivo mit H. capsulatum sensibilisiert worden waren.
