Graded doses of secretin and pentagastrin were given, on separate days, either alone or combined with a fixed dose of the other agent, to each of three healthy subjects. The kinetics of inhibition of pentagastrin-stimulated acid secretion by secretin was typical for competetive inhibition (Vmax unchanged, Km increased). Pepsin concentration and pepsin/acid ratio increased markedly with the dose of secretin. The pepsin output response to combination of secretin and pentagastrin was greater than the highest response obtained with pentagastrin or secretin alone, thus suggesting a potentiation between secretin and pentagastrin in stimulation of pepsin in man.
