Phosphonylated 8-Azahypoxantines as Acyclic Nucleotide Analogs

GRAPHICAL ABSTRACT

Abstract

A series of new phosphonylated 1,2,3-triazolo[4,5-d]pyrimidines (8-azahypoxanti-nes) was synthesized employing the 1,3-dipolar cycloaddition of corresponding ω-azidoalkylphosphonates and 2-cyanoacetamide followed by the pyrimidine ring closure with triethyl orthoformate. All compounds were evaluated in vitro for inhibitory activity against a broad variety of DNA and RNA viruses but none was found active below 100 μM. They were not cytostatic at 250 μM.
