Determination of a Synthetic Decapeptide in the Plasma of Monkeys and Rats by Narrowbore Highperformance Liquid Chromatography

Abstract

Analytical methodology utilizing solid-phase extraction and reversed phase narrowbore high-performance liquid chromatography is described for the specific determination of the decapeptide RWJ-47428-099 in the plasma of monkeys and rats. Assay validation included the demonstration of specificity, recoverability of added standard, stability, accuracy, and precision in both matrices. Linearity was demonstrated over the concentration range of 0.031 - 3.0 μg/mL in monkeys and 0.047 - 9.20 μg/mL in rats. This methodology was subsequently implemented in support of preclinical drug metabolism investigations.
