Embryogenèse somatique à partir d'embryons immatures de Blé tendre (Triticum aestivum L.): effet génotype

Abstract

Plants regeneration from in vitro tissue culture is very important for our programme to select drought—tolerant plants. In this study we examined the regeneration ability (including embryogenic potential) of twenty wheat (Triticum aestivum L.) cultivars to select high responding genotypes. For any cultivar, five culture characters were considered: rate of callogenesis, rate of germination, percentage of embryogenic calli, percentage of regeneration and mean number of regenerated plants/callus (MNRP). Results show a large genotypic effect in the production of embryogenic callus, the rate of regeneration and the MNRP as well. Differences have been observed between varieties and seven out of the twenty tested genotypes showed good somatic embryogenesis and regeneration; their use is practical in further programme related to in vitro selection.

Résumé

La régénération de plantes à partir de cellules et tissus in vitro est une étape primordiale pour notre programme de sélection de plantes tolérantes au stress hydrique. Dans le présent travail, vingt variétés de Blé tendre [Triticum aestivum L.) ont été, dans un premier temps, évaluées pour leur aptitude à la culture in vitro. Pour chacune d'elles, cinq paramètres ont été pris en considération: les taux de callogenèse, de germination zygotique, de cals embryogènes, de régénération et le nombre moyen de plantules par cal régénérant (NMPR). Les résultats obtenus montrent un effet génotype très marqué dans la production de cals embryogènes à partir d'embryons immatures, le pourcentage de régénération, ainsi que le nombre moyen de plantules obtenues par cal régénérant. Parmi les vingt variétés testées, sept seulement ont montré des taux relativement élevés de cals embryogènes et de régénération adéquats pour être utilisées dans des programmes ultérieurs de sélection in vitro.
