Bioconversion of Sugarcane Molasses into Bioplastic (Polyhydroxybutyrate) using Bacillus cereus 2156 under Statistically Optimized Culture Conditions

Abstract

The polyhydroxybutyrate (PHB) macromolecule production using cheap substrate is need of today’s life since rate of accumulate of plastic in the environment is million tonnes per year, leading to life threatening pollution problems. In this account, Response Surface Methodology (RSM) was implemented for optimization of low cost substrate (molasses) and method parameters for the manufacturing of PHB granules using Bacillus cereus and further refurbishing it into bioplastic. Primary screening of PHB manufacturing was done using Sudan black staining. 67.4 % of PHB yield of dry weight with the predicted percentage yield of 68.78 % was obtained using Box Behnken Design. Present results demonstrated that carbon source and nitrogen source were found to be the critical control factors that affected the PHB production. FTIR was performed for their functional group determination. The existing work demonstrates that produced PHB using low cost molasses is eco-friendly, economically feasible, easily available and promising alternative substrate for commercial production.
