The complete mitochondrial genome of Acheta domesticu

Abstract

The complete mitogenome of Acheta domesticu is 16,423 bp in length, contains the typical set of 37 genes, including 13 protein-coding genes (PCGs) (ATP6, ATP8, COI-III, ND1-6, ND4L, and Cyt b), two rRNAs (12S rRNA and 16S rRNA), 22 tRNAs, and a putative CR (D-loop). All PCGs start with an ATG codon except ATP8, ND3, and ND5 start with ATT and ND6 start with ATC. TAA is the most frequent stop codon although COIII end with the single nucleotide T-. The new mtDNA sequence contains 12S rRNA and 16 rRNA of rRNAs, which are located between tRNALeu and D-loop, separated by tRNAVal. The phylogenetic tree support A. domesticu has close relative with Loxoblemmus equestris.

Acheta domesticu is known under the common name house cricket, a cricket most likely native to Southwestern Asia. It is quite distinct from the other species placed in the genus Acheta, and the genus may be restricted to this one species.

The sample of A. domesticu was collected from Hefei City (117°25'07.10''E, 31°27'06.02''N), Anhui Province in June, 2018. The sample was stored in the Biology Specimen Room of the College of Life Sciences, Anhui Medical University, China (Sample code is AMHU-XS20181103). The DNA sample was stored in the −80 °C in the Cancer Cell Biology Laboratory, College of Life Sciences, Anhui Medical University. The complete mtDNA sequence of Acheta domesticu has been assigned GeneBank accession number MK204368.

The length of the complete mtDNA sequence is 16,423 bp, similar to other Gryllidae species (Cha et al. Citation2007). It contains the typical set of 37 genes, including 13 protein-coding genes (PGCs) (ATP6, ATP8, COI-III, ND1-6, ND4L, and Cyt b), two rRNAs (12S rRNA and 16S rRNA), 22 tRNAs and a putative CR (D-loop). The overall base composition for the mtDNA sequence is as follows: A, 40.4%, C, 17.5%, G, 9.5%, and T, 32.6%. The new sequence has higher A + T content (73.0%) and lower C + G (27.0%), similar to other invertebrate species (Fenn et al. Citation2007; Cha et al. Citation2007; Yang et al. Citation2016). Through the 13 PGCs, the longest one is ND5 (1720 bp), and the shortest is ATP8 (161 bp), similar with the other species (Fenn et al. Citation2007; Cha et al. Citation2007; Yang et al. Citation2016). All protein-coding genes start with an ATG codon except ATP8, ND3, and ND5 start with ATT and ND6 start with ATC. TAA is the most frequent stop codon although COIII and ND1 end with the single nucleotide T–, and ND4 stop with TA–. The new mtDNA sequence contains 12S rRNA and 16 rRNA of rRNAs, which are located between tRNALeu and D-loop, separated by tRNAVal. The 12S rRNA is 768 bp long and the 16S rRNA is 1306 bp in length. All tRNA genes posses the typical clover leaf secondary structure except for tRNASer(AGN) and tRNASer(CUN), which lacks a dihydroxyuridine (DHU) arm. The non-coding regions include a control region (D-loop) and a few intergenic spacers. The D-loop is located between 16S rRNA and tRNAVal and is 1,766 bp long.

Phylogenetic trees were estimated using ML methods, based on the complete mtDNA of six crickets, and corresponding Locusta migratoria (NC_001712) sequence was used as outgroup, sharing similar topologies and high node support values (). Chiromantes haematochir has close relative with Loxoblemmus equestris.
