ENHANCED RESONANCE LIGHT SCATTERING OF ALCIAN BLUE 8GX AS AN ASSAY OF DNA

Abstract

A novel assay of DNA with the sensitivity at nanogram levels was established in the present paper based on the measurement of enhanced resonance light scattering (RLS). In neutral aqueous solution, the interaction of ABGX with DNA results in strong enhanced RLS signals characterized by three peaks at 398.8 nm, 451.1nm and 469.0 nm. Electrostatic attraction mechanism of ABGX with DNA was established based on the measurements of RLS and absorption data. With the enhanced RLS signals at 398.8 nm, which is the strongest of the three enhanced RLS peaks, calf thymus DNA (ctDNA) and fish sperm DNA (fsDNA) in the range of 0.25 ∼ 2.0 μg/ml can be determined if 1.0 × 10−5mol/L ABGX was employed. The limits of determination were 7.1 ng/ml for ctDNA and 10.2 ng/ml for fsDNA respectively. Four synthetic samples containing ctDNA and fsDNA were determined with good reproducibility.
