Use of a Ferrous Oxidation-Xylenol Orange (FOX) Assay to Determine Lipoxygenase Activity in Barley and Malt

Abstract

The objective of this research work was to develop a new high-throughput screening method for lipoxygenase (LOX) activity, which was based on the determination of lipid hydroperoxides by ferrous oxidation-xylenol orange (FOX) assay. The final method employed extraction of a finely ground sample at a grist/buffer ratio of 1:20 for 30 min at 4°C. Extracts were incubated with linoleic acid for 5 min at 25°C, and the hydroperoxides formed were determined with the FOX reagent. Up to 100 samples can be analyzed per day with this method. The new method showed good reproducibility (coefficient of variation = 1.4–6.5%) and accuracy, although there are some disadvantages and limitations compared with the traditional UV assay. The FOX assay principal may be useful to barley breeders for screening large numbers of progeny, and also would have value to maltsters and brewers when conducting research work on LOX or performing routine quality assurance.

RESUMEN

El objetivo de la investigación fue desarrollar un nuevo método de alto rendimiento de la detección de la actividad de lipoxigenasa (LOX), que se basa en la determinación de hidroperóxidos de lípidos por oxidación ferroso-xilenol naranja (FOX) ensayo. El método final empleó extracción de una muestra molida finamente en una proporción de grano-amortiguar de 1:20 durante 30 min a 4°C. Los extractos se incubaron con ácido linoleico durante 5 min a 25°C, y los hidroperóxidos formados fueron determinados con el reactivo FOX. Hasta 100 muestras pueden ser analizadas por día con este método. El nuevo método mostró una buena reproducibilidad (CV, 1.4–6.5%) y la precisión, aunque existen algunas desventajas y limitaciones en comparación con el ensayo de UV tradicionales. El principal ensayo FOX puede ser útil para los criadores de cebada para el cribado de un gran número de progenie, y también tienen valor para los fabricantes de malteros y cerveceros cuando se realizan trabajos de investigación sobre LOX o realizar rutinario de garantía de calidad.
