A Comparison of Immediate and Delayed Acetone Fixation for Demonstration of Lymphocyte Markers in Frozen Sections

Abstract

Acetone fixation for frozen tissue sections of lymphoid tissues is commonly used for lymphocyte marker studies. This study demonstrated that for optimal reactivity, acetone fixation should be handled differently for each marker. We found that immediate acetone fixation was best for T cell markers and Leul4. Immunoglobulins were best demonstrated by drying the tissue sections in a desiccator and delaying fixation until the following day.
