Abstract
A survey was conducted with 25 formalin-fixed, aciddecalcified, paraffin-processed bone marrow trephine biopsies (BMTBs) to demonstrate mRNA of kappa and lambda light chains with dual chromogenic In Situ hybridization. The BMTBs were fixed in 8% phosphate-buffered formaldehyde and then decalcified with a commercially available combination formaldehyde/formic acid mixture. Two color (dual) chromogenic in situ hybridization was performed with the use of a commercial kit from Zytovision® (GmbH, Bremerhaven, Germany). The results showed that the histotechnical protocol of formaldehdye fixation and combined fixative/decalcifer (formaldehyde/formic acid) preserved detectable mRNA of kappa and lambda light chains in paraffin-embedded BMTBs.
