Abstract
Autoradiography increases the difficulty of good definition of nuclear details which are near the limit of visibility in usual conditions of examination. However, the material can be fully studied by two detailed microscopical examinations, the first one being done before, the second after the autoradiographic process. For the first examination, Feulgen staining followed by counterstaining with a saturated solution of Fast green in n-butyl alcohol is used; for the second, Groat's iron hematoxylin followed by 0.1% Fast green in 80% ethanol. This procedure allows a comparison to be made between a complete caryological picture and its corresponding autoradiograph.
