- During our initial PMEA synthesis according to Holy's procedure,1a we experienced problem with the purification of PMEA diethyl ester. The reported crystallization protocol could not be reproduced in our hand. We speculated that the unreacted adenine and the presence of N-7 isomer could be hindering the product crystallization, which prompted us to look for alternative PMEA synthesis
