• It is possible that the insertion of the 58 bp palindrom in locations other than that of pAS807 may also be unstable. If for example the incidence of palindrom instability in other locations is 10−5instead of IO−4 per round of plasmid DNA replication, we would have to increase either the number of clones to be analyzed from 20–30 to 200–300 clones or to increase the number of generations by a factor of 10. The latter approach is now being used to test all the palindrom insertions considered to be stable
