- Homogenisation of the leaves was done in a blender run at a low speed 3 × 20s with the following buffer: 0.5 M sucrose, 0.05 M Tris-Cl, 0.2 mM dithiothreitol, 0.5mM ATP, 0.5 mM, MgCl2. The radio-labelled precursor was added to the supernatant obtained after filtration through 4 layers of cheese cloth. The incubation was done from one day upto three weeks
- 10 uCi of [2-14C]MVA (53mCi mmol−1) and [2-14C]Acetate (1.85–2.2 GBq mmol−1; 50–60 mCi mmol−1) were fed to the plant twigs/seed kernel extracts. % Incorporation = Total radioactivity (dpm) in pure compound (Azadirachtin) × 100/Total radioactivity (dpm) administered through various methods % Incorporation of [2-14C] acetate was less and hence is not described in detail here

- Radio-active salanin was isolated from the seed kernels which were fed [2-14C] MVA and 0.5 uCi of radioactive salanin was incubated in a cell-free system prepared from fresh seed kernels. Maximum amount of radioactivity (8860 dpm) was obtained in azadirachtin isolated after a week of incubation. That shows about 0.8% incorporation of salanin into azadirachtin
