- The melting experiments were carried out in a medium salt buffer, 1 mM EDTA, 20 mM Na2HPO4, 140 mM NaCl, pH 7.2. The increase in absorbance at 260 nm as a function of time was recorded while the temperature was raised lineary from 10–80 °C with a rate of 1 °C per minute.
