Note Added in Proof: Since the submission of this manuscript an article [R. F. Kaiko and C. E. Inturrisi, Clin. Pharmacol. Ther., 18:96-103 (1975)] has appeared on the disposition of LAAM in human subjects maintained on oral 40 to 60 mg dose three times per week. The mean apparent half-lives of LAAM and noracetylmethadol in plasma were reported as 7 and 48 hr, respectively, and the plasma levels of norbisacetylmethadol remained relatively constant throughout the dosing schedule.

In another recent study [H. M. Lau and G. L. Henderson, Proc. West. Pharmacol. Soc. 18:270-274 (1975)], plasma levels of LAAM and its metabolites noracetylmethadol and bisnoracetylmethadol following acute (60 mg) and chronic oral administration in man were determined. The plasma levels of the two metabolites were shown to accumulate following chronic administration of LAAM. Amer. J. Drug & Alcohol Abuse, 2(34), pp. 307-315 (1975)
