Note Added in Proof: Since the submission of this manuscript an article [R. F. Kaiko and C. E. Inturrisi, Clin. Pharmacol. Ther., 18:96-103 (1975)] has appeared on the disposition of LAAM in human subjects maintained on oral 40 to 60 mg dose three times per week. The mean apparent half-lives of LAAM and noracetylmethadol in plasma were reported as 7 and 48 hr, respectively, and the plasma levels of norbisacetylmethadol remained relatively constant throughout the dosing schedule.
In another recent study [H. M. Lau and G. L. Henderson, Proc. West. Pharmacol. Soc. 18:270-274 (1975)], plasma levels of LAAM and its metabolites noracetylmethadol and bisnoracetylmethadol following acute (60 mg) and chronic oral administration in man were determined.
