(A). HTT facilitates an SQSTM1 droplet to properly recognize ubiquitinated cargoes and mediate autophagosome formation, resulting in complete engulfment of the droplet. (B). mHTT monomers interfere with SQSTM1-LC3 interaction on the surface of an SQSTM1 droplet; mHTT oligomers affect the properties of the SQSTM1 droplet; alone or together, mHTT changes the key factors for proper phagophore membrane bending and leads to the formation of an autophagosome without substrates.
