Abstract
Nano-delivery systems represent an astoundingly approach to the controlled delivery of therapeutic agents to promote wound regeneration. Chitosan (CS) and hyaluronic acid (HA), two oppositely charged polysaccharides, hold great promise for tissue engineering applications. In this study, we developed a polyelectrolyte complex by electrospinning CS and subsequent coating by HA, resveratrol (RS) and adipose derived stem cell (AD-MSC). The physicochemical properties of the developed nanofibrous mats were investigated. The CS/HA/RS scaffold with AD-MSC was used as a wound dressings in male rat skin wound model. Human dermal fibroblasts showed over 80% viability on the CS/HA/RS scaffold and also all scaffolds demonstrated cytocompatibility with fibroblasts. The coated nanofibers presented controlled release of HA and RS for up to 72 hours. The in vivo results revealed that CS/HA/RS with AD-MSC had a profound effect on the healing progress by augmenting wound tissue granulation and promoting wound proliferation and re-epithelialization. Thus, the current study reveals that the electrospun CS/HA/RS with AD-MSC scaffold could be proposed as an appropriate wound dressing in the in vivo wound healing.

An HA/RS/AD-MSC coated electrospun CS mats were proposed.
In vitro, fibroblasts adhered and proliferated better on CS/HA/RS scaffold.
In vivo, CS/HA/RS/AD-MSC scaffold integrated better within the wound.
