Influence of Polymeric Blend of Carbomer-Gum on the Targeted Delivery of 5-FU

Abstract

In this study, the effect of polymeric blend of Carbopol71GNF: guar gum (4:1) on in vitro release of 5-FU was studied. Premature release in upper GIT was prevented by coating the matrices with Eudragit-S100 (6.0% w/v). Korsermeyer-Peppas was the best fit model indicating Non-fickian release. Swelling studies were carried out to support the release mechanism. Stability studies were performed at 40°C/75% RH for 3 months. In vitro cecal study revealed that complete drug release would occur in the human colonic microenvironment. Therefore, it results in site-specific delivery of a hydrophilic drug for the treatment of local and systemic colonic pathologies.

Notes

It was observed that tablets containing a higher concentration of guar gum (>180 mg) alone could not be prepared by direct compression.
