- The TPP/DEAD mediated alkylation of O-substituted hydroxamates with alcohols is well documented by the work of Miller, who reported that competitive O-alkylation of the nucleophile is usually suppressed by the use of O-benzyl-N-alkoxycarbonyl-hydroxylamine derivatives.17 More recently hydroxylamine derivatives with identical N- and O-alkoxycarbonyl protective groups have been preferred,18,19 thus simplifying the synthesis of the reagent.

- Application of our protocol given in Reference 14 for the conversion of 11b to 8b (peracetylation with NaHCO3/Ac2O in CH2Cl2) to homologous trifluoroacetates 11 resulted occasionally in the competitive formation of O-monoacetylated derivatives (IR: CO at ≈ 1740 cm−1), even at prolonged reaction times. These byproducts, which exhibit very similar polarity to compounds 8 upon TLC, have been avoided by further acylation with Ac2O/pyridine.
- Prolonged reaction times with NH3MeOH, and deprotection with NaOCH3/MeOH leaded to side reactions.
