Influence of separation conditions on antimicrobial activity of polyphenolic fractions from murta leaves extract

Abstract

Response surface methodology (RSM) was employed to evaluate the effect of flow (2, 2.5 and 3 mL/min) and gradient of elution (A: water; B: methanol, step 35–45–55% B, 40–50–60% B and 45–55–65% B) on polyphenols separation from organic-aqueous murta leaves extract by Sephadex LH-20 chromatography. Six fractions were collected from extracts to which response was measured as total polyphenols and antimicrobial activity against Staphylococcus aureus. The models obtained showed that variables are significant for the inhibition activity of fraction 6 against S. aureus (p ≤ 0.05). The best separation condition of this fraction was obtained in the area where the flow is 2.5 mL/min (medium level) and the mobile phase contains a lower methanol percentage. The inhibitory activity of fraction 6 was explained by the flavan-3-ol compounds of lower molecular weight, not present in other fractions.

Mediante metodología de superficie de respuesta se evaluó el efecto de las variables flujo (2, 2,5 y 3 mL/min) y gradiente de elución (A: agua; B: metanol, escalón 35–45–55% B, 40–50–60% B y 45–55–65% B) sobre la separación de polifenoles del extracto orgánico-acuoso de hojas de murta por cromatografía en Sephadex LH-20. Los extractos eluidos fueron colectados en seis fracciones, a las que como respuesta se les midió polifenoles totales y actividad antimicrobiana frente a Staphylococcus aureus. Los modelos obtenidos demostraron que las variables estudiadas son significativas para la actividad inhibitoria de la fracción 6 sobre S. aureus (p ≤ 0,05). La mejor condición de separación de esta fracción resultó en el área donde el flujo es 2,5 mL/min (nivel central) y la fase móvil contiene un menor porcentaje de metanol. La actividad inhibitoria de la fracción 6 estaría explicada por los compuestos flavan-3-ol de menor peso molecular, no presentes en las fracciones restantes.
